“Tumor Reversion” – Re-educating Cancer: Studies Led by Professor Mariano Bizzarri

In recent years, a new frontier in oncology has opened up promising new hopes: “Tumor Reversion,” the possibility to reprogram tumor cells to revert to a normal or less aggressive phenotype, rather than eliminating them with cytotoxic approaches.

An innovative approach: cancer as disorganized tissue

Unlike traditional approaches that view the tumor as an external enemy to be destroyed, “Tumor Reversion” interprets it as disorganized, but recoverable tissue. Tumor cells are characterized by high phenotypic plasticity and can, under certain conditions, spontaneously regress or be guided toward a process of re-differentiation.

The contribution of Professor Mariano Bizzarri

Professor Mariano Bizzarri, from the University La Sapienza of Rome and head of the Systems Biology Group Lab, is among the main promoters of this innovative approach in Italy. His studies demonstrate how embryonic extracts (from zebrafish) can modulate epigenetic signals (especially the TCTP protein), inducing positive regulation of morphogenesis and regression of the malignant phenotype. Tumor cells treated in vitro and in animal models showed significant reductions in proliferation, metastatic ability, and a partial recovery of epithelial characteristics.

Biological and epigenetic foundations

“Tumor Reversion” is based on the modulation of epigenetic mechanisms without altering the genome. Tumor differentiation (acquisition of stem-like features) can be reversed by acting on transcription factors, microRNAs, and environmental signals (ECM, tumor microenvironment).

Key targets include TCTP, presenilin-1, and signaling pathways involved in the epithelial-mesenchymal transition (EMT/MET).

Clinical implications

  • Reduction of side effects related to chemotherapy and radiotherapy.
  • Overcoming drug resistance by restoring cellular homeostasis.
  • Potential integration with immunotherapy and other targeted therapies.

Limits and future perspectives

Despite promising preclinical results, controlled clinical studies are still needed to confirm efficacy, safety, and durability of response. The main risk is the unstable reversibility of the process, with possible return to the malignant phenotype. Nevertheless, “Tumor Reversion” represents a potentially revolutionary approach, focused not on destruction but on functional rehabilitation of tumor tissue.
